Tsonga, who lost 7-6 (7/5), 6-7 (5/7), 7-5, 7-6 (7/5) on Court One, cut a frustrated figure as he struggled to make any impression on the Karlovic serve.
The 6ft 10in 22nd seed, who reached the fourth round in 2004, racked up 46 aces and a phenomenal first-serve percentage of 77 per cent as he set up a fourth-round meeting with Fernando Verdasco.
Serbian fourth seed Novak Djokovic also reached the Wimbledon last 16 with a 6-4, 6-4, 6-4 win over America's Mardy Fish.
